Niraparib toegevoegd aan abirateron en prednison bij gemetastaseerd castratieresistent prostaatcarcinoom
In de hier besproken MAGNITUDE-studie wordt bij patiënten met mCRPC en een tumor- of kiembaan-BRCA1/2-mutatie na een follow-up van mediaan 18,6 maanden een statistisch significant langere rPFS gezien na palliatieve behandeling met niraparib, abirateron en prednison dan na placebo met abirateron en prednison (HR: 0,53 [95%-BI: 0,36-0,79]; p = 0,001). Dit resultaat voldoet aan de criteria voor een positief advies volgens de PASKWIL2023-criteria voor palliatieve behandeling bij studies waarin de mediane overleving in de controlegroep langer dan 12 maanden is.
