Sotorasib als tweede- of hogerelijnsbehandeling van het irresectabel of gemetastaseerd niet-kleincellig longcarcinoom (NSCLC) met een KRAS-G12C-mutatie
In de hier besproken CodeBreaK 200-studie werd een verlenging van de PFS gezien van sotorasib ten opzichte van docetaxel bij patiënten met een irresectabel of gemetastaseerd NCSLC met een KRAS-G12C-mutatie (5,6 versus 4,5 maanden, HR: 0,66 [95%-BI: 0,51-0,86]; p = 0,0017) na eerdere behandeling met platinumbevattende chemotherapie en immuuntherapie. De OS is niet beter in de sotorasibgroep dan in de docetaxelgroep (mediaan 10,6 versus 11,3 maanden; HR: 1,01 [95%-BI: 0,77 1,33]; p = 0,53). Hiermee voldoet deze studie niet aan de PASKWIL-2023-criteria voor palliatieve behandeling bij studies waarin de mediane overleving in de controlegroep 12 maanden of korter is.
